A CRISPR interference system for the nitrogen-fixing bacteriumAzotobacter vinelandii

Steven J. Russell,
Amanda K. Garcia,
Betül Kaçar

Abstract: A grand challenge for the next century can be found in mitigating the effects of changing climate through bioengineering solutions. Biological nitrogen fixation, the globally consequential, nitrogenase-catalyzed reduction of atmospheric nitrogen to bioavailable ammonia, is a particularly vital area of focus. Nitrogen fixation engineering relies upon extensive understanding of underlying genetics in microbial models, including the broadly utilized gammaproteobacterium,Azotobacter vinelandii(A. vinelandii). Here… Show more

“…Genomic integration of ancestral nifHDK into A. vinelandii followed Garcia et al ( 34 ), using established methods for transformation and homologous recombination ( 41 ), Genetically competent cells were prepared from the non-diazotrophic A. vinelandii parent strain, ∆nif, containing disruptions to all three nif , vnf , and anf nitrogenase gene clusters ( 42 ) by growth in Mo- and Fe-deficient BN medium. The Anc AK029 strain harbors a clean replacement of nifHDK with a kanamycin resistance cassette (KanR).…”
