A critical analysis of SARS-CoV-2 (COVID-19) complexities, emerging variants, and therapeutic interventions and vaccination strategies

Abstract: Coronavirus is a family of viruses that can cause diseases such as the common cold, severe acute respiratory syndrome (SARS), and Middle East respiratory syndrome (MERS). The universal outbreak of coronavirus disease 2019 (COVID-19) caused by SARS coronaviruses 2 (SARS-Cov-2) has become a global pandemic. The β-Coronaviruses, which caused SARS-CoV-2 (COVID-19), have spread in more than 213 countries, infected over 81 million people, and caused more than 1.79 million deaths. COVID-19 symptoms vary from mild fev… Show more

“…15 Antibodies and are the mainstays of Omicron's evidence. 16 The Omicron variant has approximately 32 mutations in the spike protein, most of which have been linked to antibody-binding sites, F I G U R E 1 Worldwide percentage share of Omicron variant implying that it would evade antibodies from earlier infections, vaccines, and several monoclonal antibodies used in diagnosis. 6 Whereas clinical assumptions of vaccine efficacy can only be made after a significant number of tract infections with clearly delineated vaccination status have occurred, initial assessments of immune evasion could be made in the lab using neutralization tests.…”

“…The ongoing coronavirus disease 2019 (COVID-19) is considered one of the world's worst pandemics, with more than 400 million cases and 5.8 million human deaths reported as of February 2022 ( 5 ). Since the beginning of the COVID-19 pandemic, the scientific community has focused intense efforts on studying the virus biology, the disease manifestations and management and its prevention ( 6 , 7 ). In a short time, the SARS-CoV-2 genome, the specificity of its overall structural organization and the atomic 3D structure of the most significant proteins were revealed ( 8 , 9 ).…”

“…The RNA-dependent RNA-polymerase (RdRp) is a key enzyme that plays an important role in replicating SARS-CoV-2, making it a promising drug target in developing antiviral therapy for COVID-19 (Imran et al, 2021;Pourkarim et al, 2022). In contrast to remdesivir and monoclonal antibodies that require intravenous administration in health care settings, molnupiravir is available for oral use because of its better oral bioavailability (Li et al, 2022;Malone and Campbell, 2021;Pouramini et al, 2022).…”
