A Critical Review of Methodologies to Detect Reactive Oxygen and Nitrogen Species Stimulated by NADPH Oxidase Enzymes: Implications in Pesticide Toxicity

Abstract: In this review, potential fluorescent probe applications for detecting reactive oxygen and nitrogen species (ROS/RNS) generated from NADPH oxidases (e.g., Nox2) and nitric oxide synthase enzymes are discussed in the context of pesticide toxicology. Identification of the specific marker products derived from the interaction between ROS/RNS and the fluorescent probes (e.g., hydroethidine and coumarin boronate) is critical. Due to the complex nature of reactions between the probes and ROS/RNS, we suggest avoiding… Show more

“…Gp91phox is the major catalytic subunit of NADPH oxidase. The NADPH oxidase homologues NOX1, NOX2, NOX3, NOX4, NOX5, DUOX1, and DUOX2 are expressed in a variety of cell types [9,10]. NOX2 and DUOX1 are expressed in T and B lymphocytes [28].…”

“…Many probes (fluorescent and chemiluminescent) have been previously employed to identify ROS, but there is still a lot of confusion in this field due to a lack of mechanistic rigor and the artifacts generated from reductive/oxidative activation of the probes themselves [8] , [9] . Most of these limitations have, however, been previously addressed [10] , [11] . Irrespective of the methodology used to detect ROS, it is clear that oxidants are involved, either as a major player or as a bystander, in the underlying biology.…”
