A Cytochrome f/b6 Complex of Five Polypeptides with Plastoquinol‐Plastocyanin‐Oxidoreductase Activity from Spinach Chloroplasts

Abstract: The isolation of a cytochromeflbs complex from spinach chloroplasts, with high yield and purity is reported. The complex consists of five polypeptides with a molecular mass of 34, 33,23.5,20 and 17.5 kDa, and contains one cytochromej; two cytochromes b6 and the Rieske Fe-S center with two non-heme irons. It does not contain plastocyanin and is almost completely devoid of chlorophyll and carotenoids, but lipid and detergent are present. It is lacking cytochrome b-559, although three of the five polypeptides see… Show more

“…Since the first isolation of an active b 6 f complex from spinach (Spinacia oleracea) chloroplasts (Hurt and Hauska, 1981), the purification strategy has been continuously improved and used for many photoautotrophic organisms ranging from cyanobacteria to green algae. Considerable progress resulted from the introduction of molecular genetic tag systems for the protein complex isolation, which reduced the purification time and with it the exposure time to detergents and other disintegrating components.…”

“…Activity of isolated complexes was assayed according to (Hurt and Hauska, 1981;Metzger et al, 1997) using 50 mM plastochinol as electron donor, 0.15 mM b 6 f complex and 20 mM cytochrome c as electron acceptor. The redox state of cytochrome c was monitored by spectrophotometry at 550 nm.…”

“…Cyt b 6 f complex is also a heme-containing protein complex and shows peroxidase activity, which was reported by Hurt and Hauska (1981) and by Lemaire et al (1986). Many peroxidases are heme-containing enzymes that efficiently catalyse substrate oxidations using H 2 O 2 .…”
