A direct effect of α‐chlorohydrin on rat epididymal spermatozoa

Abstract: The effect of alpha-chlorohydrin (alpha-CH) on rat epididymal spermatozoa was studied in vivo and in vitro. Alpha-CH was injected sc in doses of 5 and 20 mg daily for 16 days. The 20 mg dose resulted in diminished epididymal spermatozoal content (8 +/- 4 vs. 44 +/- 5 million, m +/- SE, n = 5) and motility (13 +/- 7 vs. 74 +/- 4%) as compared to saline injected-controls. Fertility rates were significantly reduced; control-100% (5/5), 5 mg - 25% (1/4), 20 mg - 0% (0/4). Alpha-CH was added to suspensions of sperm… Show more

“…The direct effect of a-chlorohydrin on the motility and fertility of rat epididymal spermatozoa in vivo has been confirmed (Chulavatnatol et al 1981;Tsang et al 1981;Paz and Homonnai 1982). Epithelial cells isolated from the caput epididymides of rats treated with a-chlorohydrin (20-73 daily doses of 6-8 mgjkg) showed a 50% decrease in the activity of p-galactosidase, when compared to those from untreated animals, but there was no change in the activities of P-D-glucuronidase or N-acetyl-P-D-glucosaminidase (Kemp and Killian 1978).…”

“…B. bengalensis receiving 60 and 70 mg/kg a-CH showed very low percentages of live and motile sperms which revealed the effect of the compound at the cauda epididymis level. Earlier, Paz & Homonnai (1982) also observed a marked decrease in sperm concentration and sperm motility following continuous administration of a-CH (20 mg daily) for 16 days to the laboratory rats. Results of their in uitro and in uiuo studies clearly showed that the fertilising capacity of the spermatozoa was greatly reduced at low motility; fertility was 0% at about 13% sperm motility.…”

“…The antifertility effects of -chlorohydrin, which reduces sperm motion, are well-known (Coppla, 1969;Samojlik and Chang, 1970;Vickery et al, 1974;Paz and Homonnai, 1982;Yamada et al, 1995). We analyzed sperm of rats administered -chlorohydrin with the CellSoft-4000 in a collaborative study (Ban et al, 2001).…”
