A fetal whole ovarian culture model for the evaluation of CrVI-induced developmental toxicity during germ cell nest breakdown

Abstract: Prenatal exposure to endocrine disrupting chemicals (EDCs), including bisphenol A, dioxin, pesticides, and cigarette smoke, has been linked to several ovarian diseases such as premature ovarian failure (POF) and early menopause in women. Hexavalent chromium (CrVI), one of the more toxic heavy metals, is widely used in more than 50 industries. As one of the world’s leading producers of Cr compounds, the U.S. is facing growing challenges in protecting human health against adverse effects of CrVI. Our recent find… Show more

“…RVT mitigated the effects of CrVI in the ovary, inhibited CrVI effects in the kidney and the liver. Our previous findings documented that CrVI increased the expression, phosphorylation, mitochondrial translocation, and stabilization of p53 in the ovary (Banu et al, 2011; Sivakumar et al, 2014; Stanley et al, 2015). CrVI-induced increase in Nqo1 expression could be one of the possible mechanisms for the activation and stabilization of p53.…”

“…Paraffin-embedded tissue sections were deparaffinized and TUNEL assay was performed as previously described (Sivakumar et al, 2014; Stanley et al, 2015). The apoptotic index (AI) was calculated as the average percentage of TUNEL-positive oocytes and granulosa cells from the ovaries at 400× magnification.…”

“…Our previous findings have documented an important role for the p53 pathway in CrVI-induced ovarian toxicity and apoptosis of the granulosa cells and the oocyte, as well as fetal ovarian germ cell death [39,61]. In order to evaluate if a similar mechanism exists in the placenta, the expression of ataxia-telangiectasia-mutated kinase (ATM), p53, and the p53-inducible proteins with apoptotic activity p27, PUMA and NOXA were evaluated.…”

“…Paraffin-embedded tissue sections were deparaffinized and TUNEL assay was performed as we described recently [60,61]. The apoptosis index (AI) was calculated as the average percentage of TUNEL-positive cells from each zone of the placenta at a magnification of 400x.…”
