A flap motif in human serine hydroxymethyltransferase is important for structural stabilization, ligand binding, and control of product release

Abstract: Edited by Ruma BanerjeeHuman cytosolic serine hydroxymethyltransferase (hcSHMT) is a promising target for anticancer chemotherapy and contains a flexible "flap motif" whose function is yet unknown. Here, using size-exclusion chromatography, analytical ultracentrifugation, small-angle X-ray scattering (SAXS), molecular dynamics (MD) simulations, and ligand-binding and enzyme-kinetic analyses, we studied the functional roles of the flap motif by comparing WT hcSHMT with a flap-deleted variant (hcSHMT/⌬flap). We … Show more

“…We find that Essex SHMT8 has robust catalytic activity (Table 1 and Fig. S5) comparable with SHMTs studied in other higher organisms (17,35,36). In stark contrast, Forrest SHMT8 is severely compromised in its primary, THF-dependent catalytic function (Table 1 and Fig.…”

“…Serine biosynthesis is under the control of three cytosolic enzymes: Phosphoglycerate dehydrogenase (PHGDH), which catalyzes the first step of NAD + -dependent oxidation of 3-phosphoglycerate (3PG) to 3-phosphohydroxypyruvate (3PHP); phosphoserine aminotransferase 1 (PSAT1), which converts 3PHP into 3-phosphoserine (3PS) by a glutamate-dependent transamination reaction; and phosphoserine phosphatase (PSPH), which catalyzes the last step of the serine synthesis [17]. Additionally, serine hydroxymethyltransferase (SHMT) is responsible for serine metabolism [18]. In particular, the mitochondrial isoform SHMT2 catalyzes the cleavage of serine with the production of glycine, essential for GSH and heme synthesis, and 5,10-methylentetrahydrofolate, a one-carbon unit carrier indispensable for several anabolic pathways that include de novo nucleotide biosynthesis [19,20].…”

“…SEC was used to determine the oligomeric state of AbHpaI as described previously [50]. Briefly, a Superdex TM 200 Increase 10/300 GL gelfiltration column equipped with an ÄKTA FPLC system (GE Healthcare, Germany) was equilibrated with buffer G at a 0.5 ml/min flow rate at 25 °C and A280 was monitored for protein elution.…”
