A/G polymorphism of matrix metalloproteinase 7 gene promoter region and cancer risk: A meta-analysis

Abstract: This meta-analysis was conducted to evaluate the effect of the matrix metalloproteinase 7 (MMP7)-181A/G polymorphism on cancer risk. Twenty-seven case-control studies were identified via a literature search through PubMed, Web of Science and China National Knowledge Infrastructure databases. Odds ratios (ORs) and corresponding 95% confidence intervals (CIs) were applied to assess the strength of the association between MMP7-181A/G polymorphism and cancer risk. The 27 studies were further assessed according to … Show more

“…Among these downstream target proteins, the cyclin D1 can regulate and control the cell transformation from G1 phase to S phase to interfere with the process of cell proliferation, by combining with cyclin-dependent kinase and activating it in G1 phase (24). MMP-7, a member of the family of matrix metalloproteinases, is not only involved in various extracellular matrix lysis, but also can damage diversified tumor-suppressor compositions on the cell surface (25). COX-2 can suppress tumor cell apoptosis, promote tumor cell proliferation, stimulate matrix metalloproteinases production and enhance tumor cell invasion and metastasis abilities (26).…”

“…16 MMP-7 not only participates in various extracellular matrix lysis, but also damages diverse tumor suppressor compositions on the cell surface. 17 Consequently, the overexpression of cyclin D1, MMP-7, and COX-2 plays an essential role in the proliferation, invasion, and metastasis of hepatoma cells. 18 …”

“…In colorectal cancer elevated MMP7 expression is a prognostic marker of overall survival [13, 14], associated with resistance to chemotherapy [14, 15], and increased metastasis to the liver [16]. In addition, mutations in the MMP7 promoter are associated with the increased risk of colorectal cancer [1719]. In the mammary gland, MMP7 is expressed in the normal ductal and lobular epithelia and elevated expression of MMP7 at the mRNA level has been observed in some breast carcinomas [8, 20].…”
