A Genome-Wide Chronological Study of Gene Expression and Two Histone Modifications, H3K4me3 and H3K9ac, during Developmental Leaf Senescence

Abstract: The genome-wide abundance of two histone modifications, H3K4me3 and H3K9ac (both associated with actively expressed genes), was monitored in Arabidopsis (Arabidopsis thaliana) leaves at different time points during developmental senescence along with expression in the form of RNA sequencing data. H3K9ac and H3K4me3 marks were highly convergent at all stages of leaf aging, but H3K4me3 marks covered nearly 2 times the gene area as H3K9ac marks. Genes with the greatest fold change in expression displayed the larg… Show more

“…This finding is in agreement with previous works in which treatments using stress related phytohormones or hormone analogs were reported to provoke changes in histone methylation at some hormone responsive genes in WT (Berr et al, 2010;Jaskiewicz et al, 2011;Kim et al, 2013). Previously, a strong quantitative correlation was described between the level of histone methylation and the level of expression upon stimulation for some developmental transitions (Brusslan et al, 2015;Engelhorn et al, 2017;You et al, 2017). Similarly, PR1 and PR2 in our work experienced the strongest H3K4 and H3K36 methylation increase and transcriptional induction upon stimulation in WT (Figure 5).…”

“…In a similar process, senescent Arabidopsis leaves showed a strong correlation between up-regulated senescent-associated genes marked with H3K4me3 and gene expression (Hinderhofer and Zentgraf, 2001; Brusslan et al, 2015). Also, in developing secondary xylem of Eucalyptus grandis , cell wall-related genes with vital roles in wood formation were found to be H3K4me3-enriched (Hussey et al, 2017).…”

“…The onset of leaf senescence is often accompanied by increased expression of senescence-associated genes ( SAGs ) and decreased expression of senescence downregulated genes ( SDGs ) (Gepstein et al, 2003; Breeze et al, 2011; Brusslan et al, 2015). Consistent with the delayed leaf senescence phenotype in hda9 , we found the downregulation of hallmark SAGs including SENESCENCE4 ( SEN4 ), SAG12, and SAG113 in hda9 (Figure 5A).…”
