A H2O2-producing glyoxal oxidase is required for filamentous growth and pathogenicity in Ustilago maydis

Abstract: In the phytopathogenic fungus Ustilago maydis the mating-type loci control the transition from yeastlike to filamentous growth required for pathogenic development. In a large REMI (restriction enzyme mediated integration) screen, non-pathogenic mutants were isolated in a haploid strain that had been engineered to be pathogenic. In one of these mutants, which showed a specific morphological phenotype, the tagged gene, glo1, was found to encode a product that is highly homologous to a glyoxal oxidase gene from t… Show more

“…BLASTP of NCBI shows CRO1 most closely related to conceptual translations from the basidiomycetes Cryptococcus neoformans genomes (e.g., gi57227801, score 607) and Ustilago maydis glo2 (gi33386644, score 519). The latter gene belongs to a family of glx-like genes recently studied by Leuthner et al (29). cro1 is located on scaffold 6 (coordinates 592442 to 594775), distantly linked to glx (coordinates 1201617 to 1203348).…”

“…Ustilago maydis glo1 (gi71013128, score 522) is the most closely related sequence in the current NCBI database. The membrane-bound Glo1 protein is involved in filamentous growth and pathogenicity of U. maydis (29). A variant cDNA clone extending the coding region by 102 amino acids also was amplified by reverse transcription-PCR (RT-PCR) from P. chrysosporium CC41 cultures and from colonized wood (GenBank DQ400693).…”

“…They are highly variable, but some functional classes have been identified. These include serine and cysteine protease inhibitors that target host proteases (1248,1249), effectors that minimize the levels of ROS (1250,1251), proteins that protect fungal cell walls against hydrolysis by plant chitinases (1252), and phytotoxic proteins. Apoplastic effectors may be involved in masking fungal detection by the plant by operating as protectors of fungal proteins (1253,1254).…”

“…The hydrogen peroxide appears to be involved in the degradation of lignin, because ligninase produced by the fungus needs this compound for its enzymatic activity [Leuthner et al, 2005].…”
