A high-throughput neutralizing assay for antibodies and sera evaluation against Epstein-Barr virus

Abstract: Background Epstein-Barr virus (EBV) is a wide-spread human herpesvirus that is highly associated with infectious mononucleosis and several malignancies. Evaluation of EBV neutralizing antibody titers is important for serological studies, vaccine development and monoclonal antibody screening. The traditional method based on antibody inhibition of EBV transformation of B cells is very time-consuming. A more practical flow cytometry-based (FCM) approach to evaluate neutralizing titers is not amena… Show more

“…Following the HCI protocol, 15 samples can be simultaneously detected in a 96-well plate, while by FC cells from every well should be collected in an individual FC tube and independently analysed. High-throughput approaches are widely applied for drug discovery campaigns and have been recently implemented in neutralising assays [ 20 , 51 , 52 ]. We modified a 96-well HCI technique for SARS-CoV-2 [ 23 ].…”

“…CNE2-EBV cells and Akata-EBV cells carrying the Akata-EBV-GFP BAC were used to produce epithelial cells-derived EBV (CNE2-EBV-GFP) and B cells-derived EBV (Akata-EBV-GFP), respectively 84 . 20 ng/mL 12-O-tetradecanoylphorbol 13-acetate (TPA; Beyotime) and 2.5 mM sodium butyrate (NaB; Sigma Aldrich) were added to induce CNE2-EBV cells for 12 h and then medium was replaced by fresh complete medium.…”
