A Highly Specific Enzyme-Linked Immunosorbent Assay for the Detection of Cry1Ac Insecticidal Crystal Protein in Transgenic WideStrike Cotton

Abstract: A highly selective enzyme-linked immunosorbent assay (ELISA) has been developed for the quantitative detection of the Cry1Ac protein expressed in transgenic cotton. Two Cry1Ac-specific monoclonal antibodies (MAb), Kbt and 158E6, were developed and selected to form a sandwich format ELISA. The MAb Kbt was used as a capture antibody, and 158E6 was conjugated with horseradish peroxidase and served as a detection antibody. The assay was optimized and validated with different cotton matrices. Tissues were extracted… Show more

“…Numerous techniques have been proposed for identification and classification of GM organisms, such as polymerase chain reaction (PCR)56, enzyme-linked immune sorbent assay (ELISA)78, microarrays910, electrophoresis1112, biosensors13, chips14, mass spectrometry15 and chromatographic protein profiles16. However, most of these methods are destructive, time consuming, costly, difficult operations, and impossible for rapid on-site measurements.…”

In addition, if the protein is known to be closely related to an endogenous protein or proteins, an understanding of the level of crossreactivity with the endogenous counterpart(s) is relevant.…”

“…A difference of 15% or more for each point on the standard curve has been applied in a recent method validation study (Shan et al, 2007). Criteria for determining whether the matrix effects are significant can vary and may depend, in part, on the expected precision of the standard curve.…”
