Paneth cells (PCs) were described over a century ago as granulated cells located at the base of small intestinal crypts, the 'crypts of Lieberkühn.' Various histochemical staining procedures were developed that identified PCs based on their distinctive granule-staining pattern. Early on, PCs were proposed to perform a specialized function other than absorption of digested nutrients, the predominant task of the small intestinal epithelium. Since then, many constituents of the PC granules have been biochemically characterized. The presence of various granule-associated antimicrobial substances and their release upon microbial challenge suggest that PCs function as specialized defense cells in the small intestine. Altered resistance to microbial infection in animal models with disrupted or augmented PC function provides further support for the host defense role of PCs. Other PC components suggest that PCs may also participate in the regulation of lumenal ionic composition, crypt development, digestion, and intestinal inflammation.

Özet: Bu araştırma, Ankara tavşanı ince bağırsağındaki Paneth hücrelerinin morfolojisini, histokimyasal özelliklerini, yerleşim bölgelerini ve sayısal dağılımını ortaya koymak amacıyla yapıldı. Çalışmada materyal olarak özel yetiştiricilerden sağlanan 10 adet sağlıklı, erişkin Ankara tavşanının duodenum, jejunum ve ileumlarından alınan doku örnekleri kullanıldı. Lieberhkühn kriptlerinde gözlenen Paneth hücreleri, bazalinde çekirdeği ve apikalindeki asidofilik granülleriyle tanındı. Granüller, Mallory'nin üçlü boyama tekniği, Phloxine-tartrazine, Alcian blue-performik asit ve Mallory'nin fosfotungstik asit-hematoksilen uygulamasına pozitif reaksiyon, Periyodik asit-Schiff (PAS), Alcian blue (pH 2.5), PAS-Alcian blue pH 2.5 boyamalarına ise negatif reaksiyon verdi. Paneth hücrelerinin ince bağırsağın tüm uzunluğu boyunca homojen bir dağılım göstermediği, hücre yoğunluğunun duodenumdan ileuma doğru gidildikçe arttığı gözlendi. İnce bağırsağın üç bölgesi arasındaki farkın istatistiksel olarak önemli olduğu (p<0.01) saptandı. Elektron mikroskobik incelemelerde bazı Paneth hücrelerinin apikal sitoplazmasında elektron yoğun ve homojen granüller, bazılarında ise elektron yoğunlukları farklı olan homojen granüller görüldü.Anahtar sözcükler: Ankara tavşanı, ince bağırsak, Paneth hücresi, Studies on the morphology, histochemistry and ultrastructure of Paneth cells in the small intestine of the Angora rabbitSummary: The present study was undertaken to determine the morphology, histochemical properties, localization and quantitative distribution of Paneth cells in the small intestine of the Angora rabbit. Tissue samples taken from the duodenum, jejunum and ileum of 10 healthy adult Angora rabbits, obtained from private breeders, constituted the material of the study. The Paneth cells, which were determined to be located within the crypts of Lieberkühn, were identified on their basally located nucleus and apically located acidophilic granules. These granules gave positive reactions with Mallory's triple staining technique as well as with the application of Phloxine-tartrazine, Alcian blue-performic acid and Mallory's phosphotungstic acid-hematoxylin method. Staining with periodic acid-Schiff (PAS), Alcian blue (pH 2.5), and PAS-Alcian blue pH 2.5 gave negative reactions. Paneth cells were determined not to display a uniform distribution throughout the small intestine and cell numbers were ascertained to show a gradual increase from the duodenum towards the ileum. The difference between the three regions of the small intestine was determined to be statistically significant (p<0.01).
