A Saccharomyces cerevisiae Mutant Defective in Saturated Fatty Acid Biosynthesis

Abstract: Abstract. The isolation and biochemical characterization of a Saccharomyces cerevisiae mutant, which grows only when emulsified myristic, palmitic, stearic, or oleic acid is added to the growth medium, is described. The mutant contains an enzymatically inactive fatty acid synthetase complex. The gene affected, preliminarily designated by the symbol fas, exhibits a 2:2 nuclear inheritance pattern. The formation of fatty acid synthetase in yeast is constitutive and not subject to repression by long chain fatty a… Show more

“…Radioisotope incorporation was measured: 5 ml of exponentially growing culture ['4C]acetate by the mutants to obtain instantaneous rates of total lipid synthesis in both fatty-acid-supplemented and unsupplemented cultures. To perform these measurements, we returned to conditions (leucine supplementation) favoring the nearly exclusive incorporation of [ (17,18) and Neurospora crassa (19) …”
