A Label-free Resonance Rayleigh Scattering Sensor for Detection of Thrombin Based on Aptamer Recognizing

Abstract: The interaction between thrombin binding aptamer (TBA) and thrombin (TB) was studied by resonance Rayleigh scattering (RRS). In neutral medium, TBA is present in a balanced form between a G-quadruplex structure and a random coil structure, and the TBA can be induced by metal ions to form a G-quadruplex structure. Upon addition of thrombin, the G-quadruplex selectively bound to TB, which resulted in enhanced resonance Rayleigh scattering. The scattering intensities increased proportionally with the concentratio… Show more

“…Resonance Rayleigh scattering (RRS), determined by using a commonly used fluorescence meter, has been applied to determine biomolecules and inorganic ions [81]. Metal NPs have RRS effects and the resonance scattering peak is correlated with the size of the NPs.…”

“…In vitro selected aptamers have been recently used as molecular recognition elements in place of antibodies in analytical sciences. [1][2][3] Ever since various in vitro selection techniques to obtain aptamers have been explored after phage display system was developed by Smith in 1985, 4 aptamers are increasingly becoming prevalent. In the phage display system and similar systems involving cells at any in vitro selection steps, the genetic diversity of displayed proteins/peptides is largely suppressed because the number of transformable DNA is far less than the measure of genetic diversity.…”

“…Therefore, the RRS spectra can be utilized to speculate molecular structure, charge distribution, and state of combination. Recently, the RRS technique has been widely applied to sensing metal ions, 8,9 biological molecules, [10][11][12] hazardous pollutants, 13,14 drug molecules, [15][16][17] dyes, [18][19][20] and so on. [21][22][23] The iron(III) cation is one of trace metal elements that play an important role in environmental and biological systems.…”
