A Large Family with Hereditary MTC: Role of RET Genetic Analysis in Differential Diagnosis Between MEN 2A and FMTC

Abstract: Germline mutations of the RET proto-oncogene cause three different cancer syndromes: multiple endocrine neoplasia type 2A (MEN 2A), multiple endocrine neoplasia type 2B (MEN 2B) and familial medullary thyroid carcinoma (FMTC). In the absence of biochemical and/or clinical evidence of pheochromocytoma and hyperparathyroidism, patients with MEN 2A disease display the same phenotype of FMTC disease, although prognosis and clinical management in both affected and unaffected familial members are quite different. We… Show more

“…Medullary thyroid carcinoma (MTC), a malignant neoplasm of parafollicular C-cells, represents about 5 -10 % of thyroid tumors, and contributes to the clinical presentation of MEN2 syndrome [4]. MTC releases calcitonin (CT) and carcinoembryonic antigen (CEA) and several other peptides [5,6].…”

“…SRIH transduces its effects by acting through five distinct G protein-coupled receptors, variably expressed in normal and neoplastic cells [2]. Although little is known about the identity of the specific receptor types mediating SRIH functions in tissues, some insights have been offered by studies utilizing partially selective receptor analogs [3].Medullary thyroid carcinoma (MTC), a malignant neoplasm of parafollicular C-cells, represents about 5 -10 % of thyroid tumors, and contributes to the clinical presentation of MEN2 syndrome [4].
