A Molecular Mechanism for mRNA Trafficking in Neuronal Dendrites

Abstract: Specific neuronal mRNAs are localized in dendrites, often concentrated in dendritic spines and spine synapses, where they are translated. The molecular mechanism of localization is mostly unknown. Here we have explored the roles of A2 response element (A2RE), a cis-acting signal for oligodendrocyte RNA trafficking, and its cognate trans-acting factor, heterogeneous nuclear ribonucleoprotein (hnRNP) A2, in neurons. Fluorescently labeled chimeric RNAs containing A2RE were microinjected into hippocampal neurons, … Show more

“…For example, located in the 3Ј UTR of some mRNAs are AU-rich elements (AREs), sequences involved in regulating the decay of the transcript (36)(37)(38). The interaction of hnRNP A2 with AREs in some dendritically localized mRNAs (39,40) is thought to be critical to the peripheral transport of these mRNAs (40,41). ank mRNA, which can be dendritically localized (Fig.…”

“…Previous studies in oligodendrocytes (Munro et al, 1999) and neurons (Shan et al, 2003) showed that reducing hnRNP A2 expression by using antisense oligonucleotides inhibited dendritic targeting of A2RE RNA. However, these experiments do not prove that hnRNP A2 is directly required as a trans-acting factor for dendritic targeting.…”

“…The same pathway has been shown to target exogenous heterologous RNAs to dendrites in neurons, but endogenous neuronal RNA substrates for the A2 pathway have not been identified experimentally (Shan et al, 2003). The fundamental trafficking intermediate in the A2 pathway is the RNA granule (Ainger et al, 1993), a composite structure containing multiple RNA molecules, cognate RNA binding proteins (Mouland et al, 2001), components of the translational machinery (Barbarese et al, 1995), and molecular motors for transport along microtubules .…”

“…HnRNP A2 binds to a specific cis-acting element, hnRNP A2 response element (A2RE), found in several dendritically localized mRNAs Munro et al, 1999). This interaction is necessary for transport of A2RE-mRNAs to the periphery of neural cells (Munro et al, 1999;Shan et al, 2003).…”

This interaction is necessary for transport of A2RE-mRNAs to the periphery of neural cells (Munro et al, 1999;Shan et al, 2003).A2RE-mRNAs, hnRNP A2, and other components form complexes that look by light microscopy like granules that are transported to the cell periphery by a microtubule-and kinesin-based mechanism. In cultured oligodendrocytes and neurons, granules containing A2RE-mRNAs and hnRNP A2 are associated with the cytoskeleton (CSK) .…”
