A Monoclonal Antibody to a Carbohydrate Epitope Expressed on Glycolipid and onα3β1Integrin on Human Esophageal Carcinoma

Abstract: A mouse monoclonal antibody (MAb-9) produced by immunization with a human esophageal carcinoma cell line, TE-2 (derived from undifferentiated squamous cell carcinoma) reacted specifically with about 30% of esophageal carcinoma cell lines and tissue sections from clinical samples. MAb-9 showed minimal reactivity with normal esophageal tissue. (125)I, fluorescent or gold particle labeled MAb-9 bound to TE-2 cell surfaces. (125)I-radiolabeled MAb-9 was used to detect reactive material from cell extracts in Wester… Show more

“…Human ESCC lines (KY-5, KY-10, TE-1, TE-8, YES-1, and YES-2) were provided from various sources 41-44, and they were previously characterized immunologically and morphologically in our laboratory 28, 45 Cells were grown in a medium designated as Complete medium that consisted of Dulbecco's modified Eagle medium (DMEM) containing penicillin (100 units/ml), streptomycin (100 µg/ml), L-glutamine (2 mM) (Life Technologies, Grand Island, NY), and 10% fetal bovine serum (FBS, Summit Biotechnology, Ft. Collins, CO). Cells were grown in 100-mm tissue culture dishes (Falcon, Lincoln Park, NJ) at 37°C in 100% humidity and 5% CO 2 .…”
