A multilevel investigation to discover why Kandelia candel thrives in high salinity

Abstract: The halophilic mangrove species Kandelia candel is an excellent model for understanding why halophytes thrive in high salinity. Preliminary transcriptomic analyses revealed that genes involved in diverse functions, such as in phenylpropanoid and amino acid metabolisms, and those in DNA replication and damage repair were highly responsive to salt stress. Proteomic analyses revealed that the proteins involved in light reaction of photosynthesis, amino acid and carbohydrate metabolisms, secondary metabolite biosy… Show more

“…Total RNA from the K. candel roots was also extracted using the CTAB-LiCl precipitation method (Jordon et al, 2015). Twelve DEGs were selected and analyzed by qRT-PCR using the housekeeping gene Actin as the internal reference gene (Wang et al, 2016). The primers which were designed according to the sequences obtained from transcriptome sequencing data were listed in Table 1 (see also Table S1).…”

“…GABA and Pro can be used as osmotic regulators to regulate the balance of plant internal environment (Akçay et al, 2012). Wang et al (2016) found that the contents of Glu and GABA in K. candel leaves increased significantly, but the contents of Pro did not change obviously under salt stress. In the present study, both P5CS and PDH2 genes were upregulated, which could explain why Pro was not accumulated with salt treatments in K. candel roots.…”

“…Hoagland nutrient solution was added to the sand culture. The lost water was supplemented every evening, and the nutrient solution was replaced every three days (Wang et al, 2016b). Until the plant grew four leaves (60 days), the seedlings were irrigated with Hoagland nutrient solution without NaCl (control group) and 600 mmol L -1 NaCl.…”
