A nano-sized Cu-MOF with high peroxidase-like activity and its potential application in colorimetric detection of H2O2 and glucose

Abstract: A Cu-MOF with high peroxidase-like activity was prepared and could be used for colorimetric detection of H2O2 and glucose with high selectivity and good linear range (50–500 μM).

“…To explore the cascade catalytic activity of GOx/CDs@MOF NFs, their peroxidase-like property was first investigated by the TMB chromogenic reaction. As shown in Figure A, only the mixed solution of GOx/CDs@MOF NFs, H 2 O 2 , and TMB exhibited an obvious absorption peak at 652 nm, which means that GOx/CDs@MOF NFs inherited the same peroxidase-like property of Cu-MOF. , However, the peroxidase-like activity of GOx/CDs@MOF NFs appeared under acidic conditions and showed the high activity at pH = 3–4 (Figure B), which severely hindered its applications in near neutral wound microenvironment caused by bacterial infection. Fortunately, as a natural and non-toxic catalytic substrate, the glucose can be specifically catalyzed by GOx provided by GOx/CDs@MOF NFs to reduce the localized pH value (from gluconic acid).…”

“…The peroxidase-like activities of the Ce-SrMOFs-10 were determined by observing the oxidation of TMB in the presence of the Ce-SrMOFs as nanocatalysts; the oxidation process produces a blue color with a maximum absorbance at 652 nm. 45 In brief, 100 μL of 3 mg mL −1 of Ce-SrMOFs (with the solid Ce-SrMOFs ultrasonically dispersed in an acetate buffer), 300 μL of TMB (5 mM, dissolved in DMF), and 300 μL of H 2 O 2 (30 mM) were mixed in 2.3 mL of an acetate buffer solution (200 mM, pH 4.0) and allowed to react for 30 min at 40 °C. The UV−vis spectra of the oxidized TMB product were measured between 750 and 480 nm.…”

“…For example, several Fe-containing MOFs exhibit peroxidase-like activity and some Cu-based MOFs display oxidase-mimetic catalytic ability. 17,18 Although MOFs are considered promising alternatives to natural enzymes, their catalytic activity is still lower than that of the latter. The MOF nanozymes currently studied are mainly three-dimensional block-shaped MOF crystals.…”
