Abstract. An excellent gene vector is of great importance for the development of the gene therapy. Gelatin-siloxane nanoparticles (GS NPs) with controlled size and surface charge were synthesized through a two-step sol-gel process for gene vector. In order to increase the efficiency of tumour targeting, HIV-derived Tat peptide and TTA1, a kind of modified RNA aptamer, were further grafted onto GS NPs (GS-PEG-TTA1/TAT NPs). It was demonstrated in this article that gelatin-siloxane nanoparticles (GS NPs) have the significant characteristic of being innocuous, small, covered with positive charge and even have been modified with PEG, TTA1, and TAT. The modified nanoparticles, GS-PEG-TTA1/TAT, not only have the ability of crossing the cell membrane with DNA accompanied effectively, but also could target the U251 cells and impel the DNA express right in the cells with the help of aptamer TTA1.

Background:The interleukin 28 receptor alpha (IL28RA) gene was indicated to be associated with apoptosis.However, it was not clear whether long non-coding RNA 260 (lncRNA 260)-specific siRNA targeting IL28RA gene could inhibit hypoxic reoxygenation (H/R) cardiomyocytes injury or not. To explore the mechanisms underlying the protective effects of lncRNA260-specific siRNA-mediated inhibition of IL28RA from H/R injury in cardiomyocytes, the current research was performed.Methods: The primary neonatal rat cardiomyocytes were transfected with three different pairs of siRNA specific to lncRNA260 targeting IL28RA gene and then were undergone with the conditions simulating H/R injury.Results: All three groups of cardiomyocytes treated with lncRNA260-specific siRNA experienced significantly decreased levels of lactate dehydrogenase activity and apoptosis rate relative to the nontreatment and negative control groups (P<0.05), also expressed reduced levels of IL28RA, and increased levels of PI3KCG and Bcl-2/Bax (P<0.05). Conclusions:The lncRNA260-specific siRNA may reduce cardiomyocyte apoptosis associated with H/R injury by decreasing levels of the IL28RA gene product and thus activating the PI3K/AKT signaling pathway.Keywords: Interleukin 28 receptor alpha (IL28RA); siRNA; hypoxia and reoxygenation; phosphatidylinositol 3-kinase p110 gamma (PI3KCG);

Abstract:The objective of this study was to develop angiopoietin-1 (Ang1)-expressing genetically modified human adipose tissue derived stem cells (hASCs) for myocardial therapy. For this, an efficient gene delivery system using recombinant baculovirus complexed with cell penetrating transactivating transcriptional activator TAT peptide/deoxyribonucleic acid nanoparticles (Bac-NP), through ionic interactions, was used. It was hypothesized that the hybrid Bac-NP Ang1 system can efficiently transduce hASCs and induces favorable therapeutic effects when transplanted in vivo. To evaluate this hypothesis, a rat model with acute myocardial infarction and intramyocardially transplanted Ang1-expressing hASCs (hASC-Ang1), genetically modified by Bac-NP Ang1 , was used. Ang1 is a crucial pro-angiogenic factor for vascular maturation and neovasculogenesis. The released hAng1 from hASC-Ang1 demonstrated profound mitotic and anti-apoptotic activities on endothelial cells and cardiomyocytes. The transplanted hASCAng1 group showed higher cell retention compared to hASC and control groups. A significant increase in capillary density and reduction in infarct sizes were noted in the infarcted hearts with hASC-Ang1 treatment compared to infarcted hearts treated with hASC or the untreated group. Furthermore, the hASC-Ang1 group showed significantly higher cardiac performance in echocardiography (ejection fraction 46.28% ± 6.3%, P , 0.001 versus control, n = 8) than the hASC group (36.35% ± 5.7%, P , 0.01, n = 8), 28 days post-infarction. The study identified Bac-NP complex as an advanced gene delivery vehicle for stem cells and demonstrated its potential to treat ischemic heart disease with high therapeutic index for combined stem cell-gene therapy strategy.
