The purpose of this review is to critically evaluate the available research reports on the Echinops ritro species and systematize the results obtained. This species is distributed in northern and tropical Africa, the Mediterranean basin and Central Asia. Echinops ritro has traditionally been used as medicine, mainly in China, Eastern Europe and Asia. This species contains more than 37 secondary metabolites of the species have been reported, the largest proportion of which are thiophene compounds. Phenolic compounds, flavonoids, alkaloids and lipids are also reported. Based on the discovered pharmacological and biological properties, antioxidant, antimicrobial, hepatoprotective and antitermite properties were determined. However, this species is claimed to have a wider traditional medicinal use, but its biological effect is not fully understood. Information for this study was obtained using various databases, including Scopus, Clarivate, PubMed, ScienceDirect, Google Scholar. The use of Echinops ritro in the pharmaceutical industry will help in the development of new drugs with a wide range of therapeutic activities. RezumatObiectivul studiului de față a fost evaluarea critică a rezultatelor cercetărilor efectuate asupra speciilor Echinops ritro și sistematizarea concluziilor acestora. Echinops ritro a fost utilizat în mod tradițional ca medicament, în principal în China, Europa de Est și Asia. Au fost izolați peste 37 de metaboliți secundari ai speciei, dintre care cea mai mare parte sunt tiofeni. De asemenea, specia conține compuși fenolici, flavonoide, alcaloizi și lipide. Pe baza proprietăților farmacologice și biologice identificate, s-au evidențiat proprietăți antioxidante, antimicrobiene, hepatoprotectoare și antitermite. Cu toate acestea, se consideră că această specie are o utilizare terapeutică tradițională mai largă, însă efectul său biologic nu este pe deplin înțeles. Pentru realizarea acestui studiu au fost investigate baze de date precum Scopus, Clarivate, PubMed, ScienceDirect, Google Scholar. Utilizarea Echinops ritro în industria farmaceutică ar putea contribui la dezvoltarea de noi medicamente cu o gamă largă de activități terapeutice.

Naturally, thiophenes represent a small family of natural metabolites featured by one to five thiophene rings. Numerous plant species belonging to the family Asteraceae commonly produce thiophenes. These metabolites possessed remarkable bioactivities, including antimicrobial, antiviral, anti-inflammatory, larvicidal, antioxidant, insecticidal, cytotoxic, and nematicidal properties. The current review provides an update over the past seven years for the reported natural thiophene derivatives, including their sources, biosynthesis, spectral data, and bioactivities since the last review published in 2015. Additionally, with the help of the SuperPred webserver, an AI (artificial intelligence) tool, the potential drug target for the compounds was predicted. In silico studies were conducted for Cathepsin D with thiophene derivatives, including ADMET (drug absorption/distribution/metabolism/excretion/and toxicity) properties prediction, molecular docking for the binding interaction, and molecular dynamics to evaluate the ligand–target interaction stability under simulated physiological conditions.

Momordica charantia L., or bitter melon, has been suggested to exhibit anti-inflammatory activity. In a previous study, three structurally similar triterpenes, namely 5β,19-epoxy-25-methoxycucurbita-6,23-diene-3β,19-diol (EMCD), 5β,19-epoxy-25-methoxycucurbita-6,23-dien-3β-ol (EMCO), and 5β,19-epoxy-19,25-dimethoxycucurbita-6,23-dien-3β-ol (EDMO), were isolated from bitter melon. EMCD has been shown to exhibit in vitro anti-inflammatory activity. In this study, the anti-inflammatory activities of EMCD, EMCO, and EDMO were compared. All three compounds were toxic to the RAW 264.7 macrophage cell line but not the FL83B cells. EMCD and EMCO inhibited tumor necrosis factorTNF)--induced inducible nitric oxide synthase (iNOS) expression in FL83B cells, and the IC 50 values were 19.8 and 25.7 M, respectively. By contrast, EDMO did not effectively reduce iNOS expression. Furthermore, EMCD and EMCO suppressed other TNF--induced proinflammatory signals including the activation of inhibitor kappa B kinase complex, the phosphorylation of inhibitor of nuclear factor-B, and the activation of c-Jun N-terminal kinase. EMCD consistently exhibited a higher efficacy than did EMCO in these assays. Hence, the in vivo anti-inflammatory activity of EMCD was tested. EMCD clearly repressed 12-O-tetradecanoylphorbol-13-acetate (TPA)-induced ear edema in mice. In conclusion, differences in the functional group on carbon 19 do affect the anti-inflammatory activities of EMCD, EMCO, and EDMO. EMCD exhibited the highest anti-inflammatory activity among these molecules, and its in vivo anti-inflammatory activity was confirmed.
