Anthelmintic resistance is widespread in gastrointestinal nematode populations, such that there is a consistent need to search for new anthelmintics. However, the cost of screening for new compounds is high and has a very low success rate. Using the knowledge of traditional healers from Borneo Rainforests (Sarawak, Malaysia), we have previously shown that some traditional medicinal plants are a rich source of potential new anthelmintic drug candidates. In this study, Picria fel-terrae Lour. plant extract, which has previously shown promising anthelmintic activities, was fractionated via the use of a solid phase extraction cartridge and each isolated fraction was then tested on free-living nematode Caenorhabditis elegans and the parasitic nematode Haemonchus contortus. We found that a single fraction was enriched for nematocidal activity, killing ≥90% of C. elegans adults and inhibiting the motility of exsheathed L3 of H. contortus, while having minimal cytotoxic activity in mammalian cell culture. Metabolic profiling and chemometric analysis of the effective fraction indicated medium chained fatty acids and phenolic acids were highly represented.

Abstract. The present study aimed to investigate the effect of picfeltarraenin IA (IA) on respiratory inflammation by analyzing its effect on interleukin (IL)-8 and prostaglandin E2 (PGE2) production. The expression of cyclooxygenase 2 (COX2) in human pulmonary adenocarcinoma epithelial A549 cells in culture was also examined. Human pulmonary epithelial A549 cells and the human monocytic leukemia THP-1 cell line were used in the current study. Cell viability was measured using a methylthiazol tetrazolium assay. The production of IL-8 and PGE2 was investigated using an enzyme-linked immunosorbent assay. The expression of COX2 and nuclear factor-κB (NF-κB)-p65 was examined using western blot analysis. Treatment with lipopolysaccharide (LPS; 10 µg/ml) resulted in the increased production of IL-8 and PGE2, and the increased expression of COX2 in the A549 cells. Furthermore, IA (0.1-10 µmol/l) significantly inhibited PGE2 production and COX2 expression in cells with LPS-induced IL-8, in a concentration-dependent manner. The results suggested that IA downregulates LPS-induced COX2 expression, and inhibits IL-8 and PGE2 production in pulmonary epithelial cells. Additionally, IA was observed to suppress the expression of COX2 in THP-1 cells, and also to regulate the expression of COX2 via the NF-κB pathway in the A549 cells, but not in the THP-1 cells. These results indicate that IA regulates LPS-induced cytokine release in A549 cells via the NF-κB pathway.

Objective: This study was to investigate the anthelmintic effects of ethanol extract of Curanga fel-terrae leaves on Ascaridia galli.Methods: Extract of C. fel-terrae was prepared by percolating the dried powder of the plant leaves. To observe anthelmintic effects, A. galli was exposed to the plant extract solutions in different concentrations at room temperature for 72 hrs. Sodium chloride 0.9%, sodium carboxymethyl cellulose 0.5%, and albendazole 0.1% were served as negative control, solvent control, and positive control, respectively. The anthelmintic effects were determined by observing the time taken for paralysis and the time taken for death of A. galli during experiment. Results:The results demonstrated that there were no different effects between negative and solvent controls. However, the ethanol extract of C. fel-terrae leaves exhibited significantly paralysis and death effects toward A. galli. The plant extracts at the concentration of 100 mg/ml showed similar anthelmintic effects with albendazole 0.1%. This study also indicated that the paralysis and death effects of plant extract were shorter when the worms exposed with the concentration above of 100 mg/ml. Conclusion:This study indicated that the ethanol extract of C. fel-terrae leaves has potential anthelmintic effects on A. galli.
