A new method to effectively and rapidly generate neurons from SH-SY5Y cells

Abstract: It is well known that neurons differentiated from SH-SY5Y cells can serve as cell models for neuroscience research; i.e., neurotoxicity and tolerance to morphine in vitro. To differentiate SH-SY5Y cells into neurons, RA (retinoic acid) is commonly used to produce the inductive effect. However, the percentage of neuronal cells produced from SH-SY5Y cells is low, either from the use of RA treatment alone or from the combined application of RA and other chemicals. In the current study, we used CM-hNSCs (condition… Show more

“…However, in recent years, NSC-CDM has received increasing attention due to the extensive study of the bystander behavior of NSCs in vivo, particularly that of the microvesicles released by NSCs. In addition, NSC-CDM has been shown to exert anti-apoptotic effects both in vitro [24] and in vivo [11]. To circumvent these potential obstacles and find new therapeutic strategies to treat AD, we first explored the effect of NSC-CDM in Aβ 25-35 -induced SH-SY5Y cells and we consider that NSC-CDM may be an effective treatment for AD.…”

“…To differentiate neurons, 0.5ml SH-SY5Y cells were seeded from a confluent T75 into a 6-well plate, and 10µM retinoic acid (RA) and B27 were added into the medium, which was changed every two days. Under those conditions, neurons could be obtained after one week 54 .…”

“…To obtain derivative cells with a neuronal phenotype, multiple differentiation protocols have been described [2125], but details about the final population of cells, regarding the fate-choices and fate-specification of cells that undergo terminal differentiation, have not been systematically reported. Since PD is characterized by the death of SN DAergic neurons, the degree to which this cell line displays a DAergic phenotype is a key aspect regarding the validity of the model.…”
