A new model of the disrupted latent inhibition in C57BL/6J mice after bupropion treatment

Abstract: Efficacy of rolipram but not antipsychotics to reverse the effects of bupropion suggests novel cAMP-dependent and D(2) receptor-independent mechanisms of the bupropion-induced LI deficit. Further detailed biochemical analysis of bupropion-induced LI deficit might be a fruitful approach in developing new antipsychotics.

“…Interestingly, the DISC1‐L100P variant maps to the binding domain for PDE4B (Murdoch et al 2007) and GSK‐3 β (Mao et al 2009). We found reduced interactions of DISC1 with PDE4B (Clapcote et al 2007) and GSK‐3 (Lipina et al 2010) in the brain of DISC1‐L100P mutants. In this report, we examine if the mutated DISC1 could induce alterations in DA function.…”

“…We created and characterized the DISC1‐L100P mutant mice carrying the point mutation in the second exon of the DISC1 (Clapcote et al 2007). DISC1‐L100P mice showed disruptions in prepulse inhibition (PPI) of the acoustic startle response (ASR), disrupted latent inhibition (LI), hyperactivity in the open field and impaired working memory (Clapcote et al 2007), phenotypes that are associated with DA hyperactivity (Lipina & Roder 2010; Ralph et al 2001; Shoblock et al 2003). Interestingly, the DISC1‐L100P variant maps to the binding domain for PDE4B (Murdoch et al 2007) and GSK‐3 β (Mao et al 2009).…”

“…Antidepressant or saline-treated mice were tested in the SCPP, social affiliation/novelty and the forced swim test 20 h after the last injection. The doses of drugs were chosen based on the literature (Clapcote et al, 2007;Lipina and Roder, 2010) and preliminary experiments. For the biochemical and cellular experiments, brains were collected 20 h after the last injection following either cervical dislocation or intracardial perfusion, respectively.…”

“…In their study, the cAMP-specific PDE4-I rolipram attenuated the disruptive effect of the N-methyl-D -aspartic acid (NMDA) antagonist MK-801, a commonly used deficit model in attention research, on LI in mice, thus linking PDE-Is for the first time to attention research. In another study using the same paradigm, rolipram reversed the LI deficits induced by bupropion (DA and norepinephrine (NE) reuptake inhibitor and noncompetitive nicotinic antagonist) treatment in mice (Lipina and Roder, 2009). Rolipram was also found to reverse the PCP-induced deficits in extra dimensional attentional set-shift learning (a test model for schizophrenia) (Rodefer et al, 2012).…”
