A New Take on Prion Protein Dynamics in Cellular Trafficking

Abstract: The mobility of cellular prion protein (PrPC) in specific cell membrane domains and among distinct cell compartments dictates its molecular interactions and directs its cell function. PrPC works in concert with several partners to organize signaling platforms implicated in various cellular processes. The scaffold property of PrPC is able to gather a molecular repertoire to create heterogeneous membrane domains that favor endocytic events. Dynamic trafficking of PrPC through multiple pathways, in a well-orchest… Show more

“…The cellular prion protein (PrP C ) is one of the proteins that undergoes clathrin-dependent endocytosis. [57][58][59] It has been shown to bind to Aβ oligomers as well as to p-tau. [60][61][62] Accordingly, the interaction of Aβ and/or p-tau with PrP C could be crucial for the impairment of clathrin-related vesicle endocytosis and the trafficking of vesicles.…”

“…By binding free toxic tau or α-syn oligomers in the extracellular space, exosomal PrPs prevent toxic oligomer binding to anchored PrP C and inhibit toxic signaling in the CNS of patients with diseases. Exosomes associated with PrP Sc have been shown to be infectious and pose a danger of spreading prion disease [218][219][220][221][222]. Although there is no direct study yet, exosomal PrP C might also induce CNS inflammation.…”

“…It is synthesized in endoplasmic reticulum (ER)-attached ribosomes followed by its import into ER where it is glycosylated and modified by GPI anchor before it is transported into Golgi for further modification. Then PrP C is transported to the cell surface where it can be internalized through endocytic pathway (7). The internalized PrP C can be transported into the lysosome for degradation or be enclosed in exosomes and secreted Abbreviations: Bax, Bcl-2-associated X protein; Bcl, B cell leukemia oncogene; CRC, Colorectal cancer cell; CSC, Cancer stem cell; EMT, Epithelial-Mesenchymal Transition; ERK, Extracellular-signal-regulated kinase; 5-FU, 5-fluorouracil; GBM, Glioblastomas; GPI, Glycosylphosphatidylinositol; HOP, Hsp70/90 organizing protein; HSCs, Hematopoietic stem cells; MDR, Multi-drug resistance; MET, Mesenchymal-to-epithelial transition; MMP11, Matrix metalloproteinase-11; MSCs, Mesenchymal stem cells; Oct4, Octamer-binding transcription factor 4; 1-OPRD, One octapeptide repeat deletion; PDAC, Pancreatic ductal adenocarcinoma; P-gp, P-glycoprotein; PI3K, Phosphatidylinositol 3 kinase; PrP C , Cellular prion protein; PrP Sc , Scrapie prion protein; PrP C -AptDOa, PrP C -Apt-functionalized doxorubicin-oligomer-AuNPs; SATB1, Special AT-rich sequence-binding proteins 1; SOD, Superoxide dismutase; TIMP, Tissue Inhibitor of Metalloproteinase; TNF-a, Tumor Necrosis Factor-a; TRAIL, Tumor necrosis factor-related apoptosis-inducing ligand; UPR, Unfolded protein response.…”
