A NOS1 Gene Polymorphism Associated with Asthma and Specific Immunoglobulin E Response to Mite Allergens in a Colombian Population

Abstract: Background: Nitric oxide (NO) is involved in asthma pathogenesis and is synthesized by three isoforms of NO synthase, one of them encoded by NOS1 gene. The CA-repeat and the C5266T SNP in NOS1 exon 29 have been associated with asthma and IgE levels. We thought to test the association of asthma and asthma-related phenotypes with the exon 29 CA-repeat and the C5266T SNP in a Colombian population sample. Methods: The CA-repeat and the C5266T SNP were genotyped in 167 asthmatics and 166 controls using PCR-based fr… Show more

“…In the past, measurement of small nuclear polymorphisms (SNPs) and quantitative mRNA expression by PCR technologies have been well established which can easily differentiate between highly homologous isoforms [26][27][28][29][30][31][32][33][34].…”

“…Four hundred and seventeen patients with asthma were recruited from the Social Security Clinic and public outpatient health centers between the years 2002 and 2005 in Cartagena, a tropical city on the Caribbean Coast of Colombia. Asthma was defined according to the Global Initiative for Asthma criteria, using a standardized questionnaire previously tested [26,27,28] in patients with a history of physician-diagnosed asthma. The diagnosis was confirmed by a physician belonging to the research staff, supported by a clear clinical history with clinical symptoms.…”

“…Since Blomia tropicalis and Dermatophagoides pteronyssinus are the main source of sensitization in tropical environments [33,34,35], and the prevalence of IgE sensitization to other aeroallergens is very low in asthmatics and in the general population of Cartagena [36], specific IgE against these 2 mite extracts was determined by indirect ELISA in all individuals, as described previously [28]. Absorbance was measured at 405 nm using a Spectrophotometer (Spectra Max 250, Molecular Devices, Sunnyvale, Calif., USA) and expressed as optical density (OD) units.…”

“…Blood samples were collected from all subjects for serum measurements of total serum IgE (tIgE) concentrations and DNA extraction for genotyping. Asthma definition and tIgE measurements were previously described (23)(24)(25)(26) for each population as explained in detail in the online supplement.…”
