A Novel Aeromonas popoffii Phage AerP_220 Proposed to Be a Member of a New Tolavirus Genus in the Autographiviridae Family

Abstract: Aeromonas popoffii is one of the environmental Aeromonas species. A number of factors of virulence have been described for this species and it has been reported as a causative agent of urinary tract infection. The first A. popoffii bacteriophage AerP_220 along with its host strain A. popoffii CEMTC 4062 were isolated from river water. The phage has a podovirus morphotype, shows a narrow host range and is lytic against the host strain. The AerP_220 genome comprises 45,207 bp and does not contain genes responsib… Show more

“…Genome DNA was isolated from a St 134 stock (10 9 PFU/mL) as described previously [ 36 ]. Phage particles were precipitated using a PEG/NaCl solution and suspended in a STM buffer (10 mM NaCl, 50 mM Tris-HCl, pH 8.0, 10 mM MgCl 2 ).…”

“…The inhibitory effect of isolated phage on bacterial growth was determined spectrophotometrically at OD 600 and compared with bacterial culture without phage. Furthermore, the number of revival bacteria and phage were counted following phage infection (Morozova et al 2022 ). Briefly, phage suspensions at different MOIs (0.1, 1 and 10) were co-cultured with bacterial suspensions and incubated overnight with shaking at 37 °C.…”

“…Plaques were examined after the incubation of plates overnight at 37 °C. The morphology of the phage particles was determined using transmission electron microscopy with preliminary negative staining as described previously [ 29 ].…”

“…Phage adsorption experiments were performed as described previously [ 29 ]. Briefly, the exponentially growing S. maltophilia CEMTC 2355 was infected with StenM_174 to a final concentration of 10 5 plaque-forming units per mL (pfu/mL).…”
