A novel amino acid substitution Trp574Arg in acetolactate synthase (ALS) confers broad resistance to ALS‐inhibiting herbicides in crabgrass (Digitaria sanguinalis)

Abstract: This study demonstrated that Trp574Arg substitution was the main reason for crabgrass resistance to ALS-inhibiting herbicides. To our knowledge, this is the first report of Trp574Arg substitution in a weed species, and is the first report of target-site mechanisms of herbicide resistance for crabgrass. © 2017 Society of Chemical Industry.

“…Trp 574 Arg, which was detected in only one plant of 17–56 H, had previously been found in Digitaria sanguinalis (L.) Scop. causing a broad‐spectrum resistance to nicosulfuron, imazethapyr and flumetsulam . Notably, survival rates to thifensulfuron‐methyl and imazamox of each cross‐resistant accession, i.e .…”

“…Juncus effusus, treated with the ALS inhibiting herbicide Imazamox, an Imidazolinone class herbicide, a single mutation was identified in one of the six plants sampled. This is an active site mutation, W574R (Arabidopsis numbering), W548R (Juncus numbering) previously described in the weed Digitaria sanguinalis (Li et al 2017).…”

“…Though sequences were obtained for each isolate, a mutation was only discovered in Juncus effucus acetolactate synthase. This variant is cognate to the W574R (Arabidopsis numbering) mutation previously described in Digitaria sanguinalis (Li et al 2017). However, this is the first report of the sequence of this transcript and the first discovery of a herbicide inhibiting mutant in any Poales species.…”

“…The Trp-574-Arg substitution was first reported in Digitaria sanguinalis in China [40]. Substitutions at the Trp-574 site have been documented to confer broad cross-resistance to the ALS-inhibiting chemistries.…”
