A Novel Anionic Inward Rectifier in Native Cardiac Myocytes

Abstract: Abstract-Although the cationic inward rectifiers (Kir and hyperpolarization-activated I f channels) have been well characterized in cardiac myocytes, the expression and physiological role of anionic inward rectifiers in heart are unknown. In the present study, we report the functional and molecular identification of a novel chloride (Cl Ϫ ) inward rectifier (Cl.ir) in mammalian heart. Under conditions in which cationic inward rectifier channels were blocked, membrane hyperpolarization (Ϫ40 to Ϫ140 mV) activate… Show more

“…These properties of I Cl,acid are totally or partially different from those of other cardiac Cl Ϫ currents (I Cl,CFTR , I Cl,vol , I Cl,Ca , and I Cl,ir ) identified so far. I Cl,ir (4,13,14) increases at low pH o and on cell swelling, but it inwardly rectifies. I Cl,Ca (9,11,24) requires an elevation of [Ca 2ϩ ] i for its activation.…”

“…Ϫ current (I Cl,ir ) has recently been described in cardiac myocytes (10). This current is sensitive to Cd 2ϩ and resistant to DIDS, which, with its inward rectification, distinguishes it from Cl Ϫ currents previously described in the heart.…”

“…Jentsch et al, 2002;Clark et al, 1998;Duan et al, 2000), both the muscular and the photoreceptor Cl -currents (Ugarte et al, 2005) were blocked by 9-AC with moderate efficiency. Block by extracellularly applied Zn 2+ or Cd 2+ was not observed in the current study.…”

“…Clark et al, 1998;Gründer et al, 1992;Duan et al, 2000;Rutledge et al, 2001) and represents one of the characteristics of ClC-2 currents. The present work showed a considerable augmentation of I Cl,H that might be relevant for regulatory volume decrease as supported by experiments on insect cells (Xiong et al, 1999) and Xenopus oocytes (Furukawa et al, 1998).…”
