A novel approach to investigating the erythroid lineage, using both receptor analysis and haemoglobin detection

Abstract: Progenitor cell failure in the erythroid lineage is a particular problem in bone marrow failure. To provide insight into early erythopoietic development we used sensitive techniques to examine the effects of SCF, IL-3 and MIP-1 alpha on two developmentally arrested progenitor cell lines, HEL and K562. Quantitative flowcytometric analysis showed that both expressed receptors (SCF > MIP-1 alpha > IL-3). Qualitative analysis revealed HEL cells expressed more receptors than K562 cells. Clonogenic assays with sensi… Show more

“…It has been shown that K562 cells not only expressed the receptors for mitogenic cytokines, such as SCF, IL‐3, etc. [McGuckin et al, 1996], but also expressed activin A receptors [Hilden et al, 1994, 1999]. Our results show that the activin A inhibition of mitogenic cytokine‐mediated colony formation of K562 cells is largely dependent on concentration.…”

“…More than 90% of purified cells were CD34 positive by flow cytometry. Furthermore, we included into our Ad tropism studies the cell line K562, which is considered to be an adequate model for studying gene transfer into human hematopoietic cells (44). HeLa cells, which are readily infectible by Ad5, and CHO cells, which are refractory to Ad5 infection (3), were used as positive and negative control cell lines, respectively.…”
