A novel class of fungal lipoxygenases

Abstract: Lipoxygenases (LOXs) are well-studied enzymes in plants and mammals. However, fungal LOXs are less studied. In this study, we have compared fungal LOX protein sequences to all known characterized LOXs. For this, a script was written using Shell commands to extract sequences from the NCBI database and to align the sequences obtained using Multiple Sequence Comparison by Log-Expectation. We constructed a phylogenetic tree with the use of Quicktree to visualize the relation of fungal LOXs towards other LOXs. Thes… Show more

“…Other members are 9 S -MnLOX of the rice stem root fungus, Magnaporthe salvinii ( 25 ), Mo-MnLOX of the rice blast fungus, Magnaporthe oryzae (A. Wennman et al, unpublished observation), and 13-MnLOX of Aspergillus fumigatus and homologs within Fusarium, Colletotrichum, Aspergillus, and Penicillium ( 11,12,22,23 ) ( Fig. 1B ).…”

“…Fungal LOXs with catalytic Fe, e.g., FoxLOX of F. oxysporum and the recently characterized 13-LOX of the oyster mushroom, Pleurotus ostreatus ( 26 ), form a separate subfamily with larger sequence similarities to plant and animal LOXs than to MnLOXs ( Fig. 1B ) ( 11,12 ).…”

“…This is followed by oxygen insertion, which usually produces cis-transconjugated hydroperoxy fatty acids (1,2). Plant, mammals, and a few prokaryotes express FeLOX, whereas both MnLOX and FeLOX occur in plant pathogenic fungi (5)(6)(7)(8).…”

“…All LOX belong to the same gene family, but plant FeLOX, mammalian FeLOX, and fungal FeLOX and MnLOX form separate subfamilies (5,8). The prototype MnLOX is secreted by the take-all fungus of wheat, Gaeumannomyces graminis (7).…”

“…The medium was harvested, 136 g (NH 4 ) 2 SO 4 was added per liter, the medium was adjusted with in addition to M. oryzae (9)(10)(11)(12). They are named by their catalytic properties with 18:2n-6 as a substrate or by their origin: Gaeumannomyces graminis (13R-or Gg-MnLOX), Magnaporthe salvinii (9S-or Ms-MnLOX), Aspergillus fumigatus (13-or Af-MnLOX), Fusarium oxysporum (11R/13S-or Fo-MnLOX), and Colletotrichum gloesporioides (9S/11S-or Cg-MnLOX) (2,(9)(10)(11)(12). Based on sequence homology to Gg-MnLOX, a number of LOX sequences have been denoted as putative MnLOXs at NCBI.…”
