A novel defensin‐like peptide from salivary glands of the hard tick, Haemaphysalis longicornis

Abstract: A novel defensin-like antimicrobial peptide named longicornsin was isolated from the salivary glands of the hard tick, Haemaphysalis longicornis, using a 10-kDa cut-off Centriprep filter and reversed-phase high-performance liquid chromatography (RP-HPLC). Its amino acid sequence was determined as DFGCGQGMIFMCQRRCMRLYPGSTGFCRGFRCMCDTHIPLRPPFMVG by Edman degradation. The cDNA encoding longicornsin was cloned by cDNA library screening. The predicted protein from the cDNA sequence was composed of 78 amino acids in… Show more

“…In addition, this is the first study showing the activity of tick defensins against the above microorganisms. The antibacterial activity of tick defensins has been reported to be mainly directed against Gram + bacteria, but some isoforms were reported to also have activity against Gram-bacteria [44][45][46][47][48][49]. However, the activity of the defensins tested in this study seems to be rather specific, mainly against fungi.…”

“…Early reverse genetics approaches (Figure 2B) enabled the search for specific genes by nucleic acid hybridization-based screening of cDNA libraries produced from tick salivary glands [24-26]. Protein-coding cDNAs of interest were cloned, overexpressed using various systems, and their function characterized in bioassays [27,28].…”

“…From this perspective ticks were postulated to inject anti-microbial peptides into the feeding site to prevent the feeding site from being infected [23,48]. Multiple anti-microbial peptides have been characterized in ticks, a majority of which are defensins [98][99][100][101][102][103][104][105][106], microplusin/microplusin-like [107][108][109] and hebreain/hebreain-like [110]. In this study seven of the 15 anti-microbial peptides in S2 Table and Fig 4B are microplusin-like, a single lysozyme, and the rest, are characterized by pathogen-recognition domains (n = 7).…”
