A Novel Model of Continuous Depletion of Glutathione in Mice Treated With L-Buthionine(s,r)-Sulfoximine.

Abstract: L-buthionine (S,R)-sulfoximine (BSO), an inhibitor of glutathione (GSH) synthesis, was administered to mice via drinking water for 14 days in order to establish an animal model with continuously depleted levels of GSH. No toxicity was observed at 20 mM BSO, even though a significant decrease in liver weight was observed at 30 mM BSO. GSH levels in the liver, kidney, brain, lung, heart, spleen, pancreas, small intestine, large intestine, skeletal muscle, plasma and blood cells from mice given 20 mM of BSO were … Show more

“…3l). To extend this observation, three cohorts of Tg:Pax7-nGFP mice received water or a daily administration of the glutathione donor N-acetyl-L-cysteine for 1 week to protect against ROS, or buthionine sulfoximine for 15 days to deplete glutathione within cells as previously described 35,36 . The number of satellite cells that survived 4 days post mortem was not significantly different within these three sets of mice (Fig.…”

“…To prove this, we tried to deplete the hepatic GSH level using buthionine sulfoximine et al ., 1979). Male C57BL/6J mice were administered 20 mM of BSO in drinking water for four days to deplete the GSH continuously (Watanabe et al, 2003). By this administration, the hepatic GSH level decreased about half of the control level (Fig.…”
