A novel monoclonal antibody (FUN‐1) identifies an activation antigen in cells of the B‐cell lineage and Reed—Sternberg cells

Abstract: The characterization of a new monoclonal antibody (MoAb) recognizing a human B-cell activation antigen, designated FUN-1, is described in this paper. Immunoprecipitation revealed that FUN-1 recognizes an antigen with a molecular weight (MW) of 75kD. FUN-1 reacts with pokeweed mitogen-activated B lymphocytes and monocytes of peripheral blood, but not with unstimulated lymphocytes or granulocytes. It also reacts with large lymphoid cells in germinal centres, Epstein-Barr virus-transformed B cell lines, large B-c… Show more

“…B7-1 expression on H/R-S cells was more or less similar to CD30 expression, confirming previously published data (Nozawa et al, 1993;Gruss et al, 1994;Munro et al, 1994). In addition to H/R-S cells, reactivity in normal British Joumal of Cancer (1997) 76(9) (Freeman et al, 1989;Guinan et al, 1994).…”

“…As H/R-S cells are believed to be the malignant cells in HD, these cells seem to be the best target for such immunotherapy. The antigenic make-up of H/R-S cells include several well-expressed surface antigens: CD15, CD30, CD40, IL-2R (CD25), transferrin receptor (CD71), B7-1 (CD80) and B7-2 (CD86) Durkop et al, 1992;Falini et al, 1992a;Nozawa et al, 1993;Smith et al, 1993;Gruss et al, 1994;Munro et al, 1994;Falini et al, 1995), to which a therapeutic agent could be targeted. The active moiety coupled to a MAb may consist of a radio isotope (radioimmunoconjugate) (Vriesendorp et al, 1991) or a protein synthesis-inactivating toxin (immunotoxin, IT) (Falini et al, 1992b).…”

“…CD34 is a ligand for L-selectin and is expressed by vascular endothelial cells (Ricono et al 2003;Pusztaszeri et al 2006). FB21 is a monoclonal antibody that binds a sialic-acid-dependent surface antigen and reacts with human glomerular endothelial cells (Nozawa et al 1993(Nozawa et al , 1994Kawasaki et al 2004). We previously reported that FB21 reacted with the glomerular endothelial cells of normal kidneys and was detected on the surface of glomerular endothelial cells, and noted that FB21 staining of glomerular endothelial cell was significantly weaker in patients with hemolytic uremic syndrome (HUS) and Henoch-Schoenlein purpura nephritis (HSPN) than in children with normal kidneys, and indicated that FB21 can be used as a marker for glomerular endothelial cell injury and dysfunction in HUS and HSPN patients (Kawasaki et al 2004).…”

“…The costimulation pathway includes CD70, CD72, and CD80/CD86 and their receptors CD27, CD5, and CD28, respectively. H-RS cells are usually CD54+, CD58+, CD80+, CD86+, and less frequently CD70+, but usually CD72- [23,54,74,85,89]. Direct contact between H-RS cells and T cells via MHC molecules, cell adhesion molecules and costimulator molecules could sig nificantly increase T-cell infiltration in tissues ( fig.…”

“…The expression of CD70 and CD80/CD86 (as mem brane-bound cytokines), though observed frequently in some types of NHLs, may have a unique function in HD because there is an intimate contact between H-RS cells and CD27-positive (CD27 +) or CD28 + T cells (class 4) [23,74,85,89]. CD27 and CD28 function as the cyto kine receptors for CD70 and CD80/CD86, respectively [19,51].…”
