A novel polymer‐based genosensor for the detection and quantification of Streptococcus pneumoniae in genomic DNA sample

Abstract: The Streptococcus pneumoniae detection plays an important role in the diagnosis and monitoring of pneumococcal diseases. A genosensor based on graphite electrodes modified with polymer was developed for such detection. First, the poly(4‐aminophenol) film was electrochemically deposited on a graphite electrode. Afterward, an S. pneumoniae‐specific oligonucleotide (Strep1), isolated from conserved regions of the bacterial genome, was immobilized onto the modified electrode surface and used for the test with comp… Show more

“…Ethidium bromide (EB) functions as an indicator that was efficiently intercalated into the dsDNA. Notably, the current responses generated by EB in the presence of S. pneumoniae were monitored using DPV analysis [ 143 ]. Using this process, the LOD was 54 ng mL −1 , revealing lower sensitivity than the colorimetric LAMP-based method [ 8 ].…”

“…Thereafter, a particular sequence of S. pneumoniae was fixed on the modified electrode superficies and employed to the assay with genomic DNA or complementary tDNA (Scheme 8). The achieved detection limits using EIS (electrochemical impedance spectroscopy) and differential pulse voltammetry were 28 and 54 ng/ml, respectively 120 …”

“…17 Electrochemical genosensors offer a quick tool for detection and diagnosis, based on the use of specific probe for gene of virulence, such inlA and hly gene for Listeria monocytogenes , 7,17 or oligonucleotides isolated from conserved region of the bacteria genome for Streptococcus pneumonia. 18 Infection by Streptococcus pyogenes causing damage of heart valves in human, as reported by 8 an electrochemical genosensor based on sepB gene probe can offer a rapid detection in few minutes (30 min) of the infection with a lower LOD of 0.10ng of ssG-DNA per 6μl (human throat swab sample s). Based on oligonucleotides probe that was identified from the 16s rRNA coding region of the Escherichia coli O157:H7 genome, Pandey et al 19 have coupled it with cystine flower-gold particles matrice to improve its immobilization, their genosensor exhibited a linear response with LOD of 1x10 -15 M provide a selective tool for Escherichia coli O157:H7.…”
