A Novel Role of Matrix Metalloproteinase-8 in Macrophage Differentiation and Polarization

Abstract: Background: Macrophages differentiation and/or polarization have been implicated in many inflammatory diseases. Results: Matrix metalloproteinase-8 (MMP8) promotes M2-macrophage differentiation and polarization through modulating TGF-␤ bioactivity. Conclusion: MMP8 plays a novel role in macrophage differentiation and polarization. Significance: The findings of this study significantly increased our understanding to biological functions of MMP8, the mechanisms for macrophage differentiation and polarization, an… Show more

“…For in vitro foam cell formation analyses, bone marrow mononuclear cells isolated from WT or NE_KO mice were differentiated into BMMs as described in our previous study, 23 followed by an incubation with 5 lg/mL of DiI-Ac-LDL (10574053, Fisher Scientific) for 5 hours. The cells were fixed with paraformaldehyde and counterstained with DAPI.…”

“…Real-time quantitative PCR (RT-qPCR) was performed as described previously. 8,23,29,30,36 Briefly, total RNAs containing small RNAs (miRNAs) were extracted from cells using the mirVana Protein and RNA Isolation System Kit (Thermo Fisher Scientific Inc) or TRI reagent (Sigma-Aldrich), according to the manufacturer's instructions, and subjected to DNase I (Sigma-Aldrich) digestion to remove potential DNA contamination. Reverse transcription for long RNA was performed using an Improm-II RT kit (Promega) with RNase inhibitor (Promega) and Random primers (Promega).…”

“…For example, MMP12 and MMP28, both macrophage products, either promote or restrict macrophage influx into lung (10, 11), and MMP8 promotes M2 polarization (12). In addition, we reported that MMP28 and TIMP3, an MMP inhibitor, moderate M1 activation of macrophages in models of lung infection and fibrosis (13, 14).…”
