A Novel Signal-Amplified Immunoassay for the Detection of C-Reactive Protein Using HRP-Doped Magnetic Nanoparticles as Labels with the Electrochemical Quartz Crystal Microbalance as a Detector

Abstract: A novel horseradish peroxidase- (HPR-) doped magnetic core-shell Fe3O4@SiO2@Au nanocomposites (Fe-Au MNPs) were employed on immunoassay for the determination of C-reactive protein (CRP) based on a electrochemical quartz crystal microbalance detector (EQCM). Firstly, the secondary CRP antibody and HRP were both immobilized on the Fe-Au MNPs (Fe-Au MNPs-anti-CRP2/HRP) as a signal tag. Secondly, the above tag and the primary antibody (anti-CRP1) in the bottom of 96-well microtiter plate were employed to conjugate… Show more

“…The advantages of oriented immobilization of biologically active protein G are good steric accessibilities of active binding sites of the antibody and increased stability and sensitivity of the immunosensor [13]. The obtained results were better those obtained with QCM (Quartz Crystal Microbalance) and ELISA techniques in human plasma using doped magnetic core-shell [8].…”

“…The linear range was 0.003-200 ng/mL and the detection limit was 1 pg/mL. 17 Compared to our new method demonstrated in this study, the authors used a more sophisticated procedure and required more chemicals. Their experiments also needed much longer time to finish.…”

“…Gan et al functionalized the surface of a QCM with a combination of Fe 3 O 4 and Au nanoparticles [ 104 ]. The Fe 3 O 4 magnetic nanoparticles were coated with SiO 2 , immersed in APTES to form amine groups (NH 2 ), and then coated with Au nanoparticles that were used to immobilize anti-CRP and HRP.…”
