A paper microfluidics-based fluorescent lateral flow immunoassay for point-of-care diagnostics of non-communicable diseases

Abstract: An easy-to-use, low-cost and portable fluorescent lateral flow immunoassay based on paper microfluidics was developed for the point-of-care diagnosis of non-communicable diseases.

“…Sandwich type assays for cTnI have been implemented in LFA while using detection strategies that rely on fluorophores [24], quantum dots [28], gold nanoparticles [29], or Raman tags [30], to name a few. On all of these systems, as in most LFA devices, NC is used in the analytical strip.…”

“…We further study the effect of layering CNF on the test line regions of the cellulose strip in the signals generated ( Figure 1d). As a model system, we consider the quantitative detection of cardiac troponin I (cTnI), a biomarker of myocardial cell damage, by a fluorescent LFA [24]. The system relies on the recognition of the target cTnI molecule by fluorescently labelled anti-cTnI detection antibodies and subsequent capture of the complex at the test line by an anti-cTnI capturing antibody (Figure 1b,c).…”

“…The comparison of the developed aptamer-based paper strip with previously reported assays for detection of cTnI is shown in Table S1 in the Supplementary Material . Compared with other paper-based assays, 16 , 25 , 26 the developed paper strip used an aptamer, which is more thermally stable, has better tolerance to wide range of pH, and is a lower cost recognition element ( times lower cost compared to the cost of cTnl antibodies purchased online). 34 , 35 In addition, using a portable reader, it showed good sensitivity close to the lower end of physiological range and required a shorter detection time than the other assays based on aptamers.…”

“…Recently, various POC testing systems for cTnI have been developed; 14 29 however, these systems have limitations in cost effectiveness (especially in the cost of the disposable cartridges), poor sensitivity, or limited stability of the recognition element. Paper-based sensing systems have demonstrated potential as a POC platform to implement assays due to the merits of paper (e.g., abundance, low cost, and ease of disposability).…”
