A photoinduced growth system of peptide nanofibres addressed by DNA hybridization

Abstract: Spatiotemporal control of peptide nanofibre growth was achieved by photocleavage of a DNA-conjugated β-sheet forming peptide that is linked through a photoresponsive amino acid residue. Peptide nanofibres were selectively formed by photocleaving the conjugate on a complementary DNA-immobilised glass substrate.

“…We previously reported a light-induced peptide nanofibre growth system using conjugate 1 , consisting of β-sheet-forming FKFEFKFE peptide and addressing single-stranded DNA (dA 20 ), which are linked by a photo-cleavable amino acid X (Fig. 1b ) 49 . Ultraviolet (UV) irradiation to conjugate 1 releases free FKFEFKFE peptide, which self-assembles to form peptide nanofibre.…”

“…Several Spr-containing peptides were already used to control function of peptides/ proteins in living cells. 14,16,21) Spr has been recently employed by some other groups for in-cell imaging of a protein of interest, 21) UV-induced propulsion of a giant liposome, 22) UV-triggered nanofiber formation, 23) and tumor-targeting therapy. 24) Application of Spr in topics not mentioned in this paper 14,17,18,25,26) are detailed in our recent review.…”

“…Among the external stimuli, light is particularly attractive because of the ease of manipulation with localized and on‐demand irradiation. We developed a system of light‐induced peptide nanofiber growth with spatiotemporal control by using peptide‐DNA conjugates (Figure ) . We synthesized conjugate 1 consisting of a β‐sheet‐forming FKFEFKFE peptide and an addressing single‐stranded DNA (dA 20 ), which are linked by a photo‐cleavable amino acid X (Figure a).…”

“…We developed a system of light-induced peptide nanofiber growth with spatiotemporal control by using peptide-DNA conjugates (Figure 8). [123] We synthesized conjugate 1 consisting of a b-sheet-forming FKFEFKFE peptide and an addressing single-stranded DNA (dA 20 ), which are linked by a photocleavable amino acid X (Figure 8a). Upon irradiation of UV light, conjugate 1 releases free FKFEFKFE peptide by the photocleavage reaction of X, which induces self-assembly of the peptide to form nanofibers 10-20 nm in width, as observed by TEM.…”
