A Pilot Study of Circulating miRNAs as Potential Biomarkers of Early Stage Breast Cancer

Abstract: BackgroundTo date, there are no highly sensitive and specific minimally invasive biomarkers for detection of breast cancer at an early stage. The occurrence of circulating microRNAs (miRNAs) in blood components (including serum and plasma) has been repeatedly observed in cancer patients as well as healthy controls. Because of the significance of miRNA in carcinogenesis, circulating miRNAs in blood may be unique biomarkers for early and minimally invasive diagnosis of human cancers. The objective of this pilot … Show more

“…Although it has been in argument and discussion on reference strategies for normalizing circulating miRNAs, there is still no consensus (Ma et al, 2012). 5s rRNA, U6 snRNA, and miR‐16 are frequently used as internal normalization controls (Luo et al., 2009; Pineles et al, 2007; Zhao et al., 2010). In this study, U6 snRNA showed low abundance in the plasma samples.…”

“…The plasma DNA concentration should be a marker for breast cancer screening that was easily accessible and noninvasive [16]. In addition, high sensitivity and specificity of the plasma DNA with fluorometric method provide a diagnostic advantage in screening.…”

“…As serum and other body fluids contain ribonucleases, miRNAs are thought to be shielded from degradation by being packed in lipid vesicles, in complexes with RNA-binding proteins, or both (115). In a pilot study of circulating miRNAs as potential biomarkers for early-stage breast cancer, 48 serum miRNAs were found to be differentially expressed in patients when compared with controls (116). Of these, 22 were upregulated and 26 were downregulated.…”
