A Pivotal Role for Heat Shock Protein 90 in Ewing Sarcoma Resistance to Anti-Insulin-like Growth Factor 1 Receptor Treatment: In vitro and In vivo Study

Abstract: Ewing Sarcoma (ES) shows several deregulated autocrine loops mediating cell survival and proliferation. Therefore, their blockade is a promising therapeutic approach. We previously reported the in vitro effect of insulin-like growth factor 1 receptor (IGF1R)/KIT pathway blockade on ES cell lines, and we now extend our observations to changes induced by this treatment in interacting proteins/networks. A proteomic analysis revealed that Heat Shock Protein (HSP)90 was differentially expressed between ES cell line… Show more

“…However, rather mild β-catenin induction by EWS-FLI1 in the eSZ (Supplemental Figure 9A) suggests that additional genetic events might be required for constitutive activation of the pathway. Pathways involving receptor tyrosine kinases are also important in Ewing's sarcoma (40,51), as was indicated in our model. Indeed, potential clinical benefits from the use of pazopanib, a multikinase inhibitor, for the treatment of childhood sarcoma, including Ewing's sarcoma, have been reported recently (52).…”

“…RNA extraction was performed using Qiagen RNA extraction kit, following the manufacturer's instructions, as described elsewhere (26,33). Retrotranscription was performed using 500 ng of total RNA.…”

“…16). Targeting of EWS/FLI1 by RNA interference in ES cells affected IGF1/IGF1R survival pathway (17) and its downstream targets (14). Both mitogen-activated protein kinase (MAPK) and PI3K signaling pathways are constitutively activated in ES, probably because of the presence of IGF1R-mediated autocrine loops.…”
