A Polymorphism of the Norepinephrine Transporter Gene in Bipolar Disorder and Schizophrenia: Lack of Association

Abstract: Norepinephrine is one of the neurotransmitters which has been implicated in the pathogenesis of mood disorders and schizophrenia. The norepinephrine transporter (NET) gene may be a candidate gene for the study of the genetics of these disorders. In this study, 198 patients with schizophrenia and 100 patients with bipolar disorder were analysed for a silent mutation 1287 A/G, located in the coding region (exon 9) of the NET gene, to determine the association between this polymorphism of the NET gene and bipolar… Show more

“…Several investigators have reported the relationship between the NET G1287A polymorphism and several psychiatric disorders including MDD, 7,14,15) schizophrenia, 8) alcohol dependence 10) and panic disorder 16) in Caucasian and Han Chinese populations, and none of them showed significant relationship. The present results replicated those by Owen et al, 7) Zill et al, 14) and Chang et al 15) in a Japanese population.…”

“…[8][9][10] However, its involvement in susceptibility to MDD has not yet investigated in Oriental populations. In the present study, the authors investigated whether the NET G1287A polymorphism is associated with the susceptibility to MDD first in a Japanese population.…”

“…Several genetic variants have been identified in the human norepinephrine transporter gene; however, no association has been found so far between those polymorphisms and either bipolar disorder and schizophrenia. 294 It has not yet been studied if genetic polymorphisms in the norepinephrine transporter gene have influence on antidepressant drug response especially of the NRIs.…”

“…It has been reported that NET has several genetic polymorphisms. Although a silent G1287A mutation was detected as a highly polymorphic nucleotide exchange in exon 9 of the NET gene by Stober et al [8], this polymorphism is not associated with the Inoue/Itoh/Yoshida/Shimizu/Suzuki susceptibility to major depression, schizophrenia, alcohol dependence and panic disorder in Caucasian populations [9][10][11][12]. Recently, Zill et al [13] have reported the development of a real-time PCR-based assay system for the novel T-182C polymorphism in the promoter region of the NET gene and examined the association between this polymorphism and major depression in a German population.…”
