A potential role of aminoglycoside resistance in endemic occurrence of Pseudomonas aeruginosa strains in lower airways of mechanically ventilated patients

Abstract: Pseudomonas aeruginosa isolates from a 5-bed intensive care unit were fingerprinted with 26 pulsed-field gel electrophoresis and tested for aminoglycoside resistance genes aac(6′)-Ib, aac(3″)-IIa, ant(2″)-27 Ia, armA, rmtA, and rmtB and integrons and virulence genes/operons phzI, phzII, phzM, phzS, apr, lasB, plcH, plcN, 28 pilA, algD, toxA, exoS, exoT, exoY, and exoU. Two major clusters were identified (49 and 19 isolates), harbouring 29 aac(6′)-Ib, bla PSE-1 , and ant(3″)-Ia genes or ant(2″)-Ia gene, respect… Show more

“…The population structure of P. aeruginosa is reported as multiclonal in most studies [ 27 , 28 ]; even when major clones were present, the population was characterized by high diversity and by the presence of several different clones. Similarly, multiple versatile P. aeruginosa clones may be circulating in our hospital, as reported in a previous study with a more limited scope conducted in a ward of the same hospital [ 29 ]. Apparently, the persistence of one or a few successful clones is unlikely, in contrast to A. baumannii [ 22 , 23 ].…”

“…Genetic relatedness among isolates was analysed by PFGE. Plugs were prepared as previously described (Mózes et al , 2014) and digested with Apa I. A CHEF DRIII machine (Bio-Rad) was used to separate the fragments in 1 % SeaKem Gold agarose (Lonza) at 14 °C.…”

“…Plugs were prepared as described earlier [13], macrorestriction was performed using XbaI (Fermentas, Vilnius, Lithuania) in a CHEF DRIII machine (Bio-Rad) in 1% SeaKem Gold agarose (Lonza) at 14°C. Electrophoresis was performed at 6 V/cm, with a reorientation angle of 120°, and switch times were ramped between 2 and 64 s for 20 h for both species.…”
