A Rapid SARS-CoV-2 RT-PCR Assay for Low Resource Settings

Abstract: Quantitative reverse transcription polymerase chain reaction (RT-qPCR) assay is the gold standard recommended to test for acute SARS-CoV-2 infection. However, it generally requires expensive equipment such as RNA isolation instruments and real-time PCR thermal cyclers. As a pandemic, COVID-19 has spread indiscriminately, and many low resource settings and developing countries do not have the means for fast and accurate COVID-19 detection to control the outbreak. Additionally, long assay times, in part caused b… Show more

“…To prevent the rapid spread of the virus, early diagnostics are vital for reducing the risk of further transmission. The current diagnostic approaches rely on nucleic acid detection of COVID-19, such as PCR/QPCR and next-generation sequencing [97,98]. However, these techniques have some drawbacks, including low efficiency, false negatives, expensive instruments and high-level personnel expertise.…”

“…Further to this, RNA extraction methods use large amounts of solvents that require bespoke storage and disposal. Use of D2PCR for detection of COVID-19 has been previously demonstrated 21,22,23 . Herein we present a clinically approved high-throughput methodology, developed using the Genesig® Real Time PCR COVID-19 High Throughput HT-CE kit V2.0 containing an optimised buffer formulation which overcomes sample-mediated PCR inhibition.…”

“…With a process of sequencing by synthesis, the amplified DNA was sequenced. The complete SARS-CoV-2 genome was obtained by overlapping DNA regions that were also high-abundance sequences [5,6].…”
