Although cerebellar mossy fibers are the most abundant cerebellar afferents and are deeply involved in cerebellar function, the organization of their projection has remained obscure, particularly in relation to cerebellar compartmentalization. The dorsal column nuclei (DCN) are a major source of cerebellar mossy fibers and possess distinct somatotopic representations of specific somatosensory submodalities. We reconstructed individual dextran-labeled DCN axons completely from serial sections and mapped their terminals on the longitudinal cerebellar compartments that were visualized by aldolase C immunostaining to clarify their projection pattern. Individual axons branched and formed about 100 rosette terminals in the cerebellar cortex, but infrequently projected to the cerebellar nuclei (1 out of 15 axons). Cortical terminals were clustered in multiple areas in the vermis and pars intermedia mostly, but not exclusively, ipsilateral to the origin of the axon. The gracile, cuneate, and external cuneate nuclei (ECuN) mainly projected to the copula pyramidis and lobule V, paramedian and simple lobules, and lobules I-V and VIII-IX, respectively, although there was some overlap. The majority of terminals were located within aldolase C negative or lightly positive compartments. However, terminals of a single axon can be located on aldolase C-negative as well as on aldolase C-positive compartments. In particular, the rostral ECuN, which is responsive to shoulder movements, projected consistently to lobule IX, which were mostly aldolase C-positive. In sum, DCN-cerebellar axons project to multiple compartments with terminals clustered mainly in the conventional spinocerebellar region with a coarse topography, which shows some relationship to the cortical compartments defined by aldolase C.

The present study was undertaken in order to determine the subnuclear origin of olivary parasagittal zones in the uvula of the rat cerebellum. Microinjections of horseradish peroxidase were made in different sublobules of the uvula (sublobules IXa, b, and c of Larsell, 1952) and in different mediolateral positions within these sublobules. The results suggest that different specific regions of the contralateral inferior olive project to each of the parasagittal zones found within the uvula. Injections in the region of the most medial parasagittal zone result in the retrograde labeling of cells in ''subnucleus c'' and nucleus β subdivisions of the medial accessory olive (MAO). Injections in the intermediate region of the uvula result in the labeling of cells in the dorsomedial cell column. Finally, injections in the lateral part of the uvula result in the labeling of cells in the ventral lamella of the principal olive (vPO) or the rostral part of the MAO or both. There is some suggestion that the more rostral sublobules receive their input from the rostral MAO while the more caudal sublobule receives its input primarily from the vPO. These results agree fairly well with similar analyses of this projection system in the cat by Brodal in 1976.

To assess the locations and densities of cells in the dorsal medulla giving rise to ipsilateral versus contralateral projections to the mechanosensory regions of cerebellar cortex in the anterior lobe and paramedian lobule, these cortical regions were injected unilaterally with horseradish peroxidase in each of 5 raccoons. To show injection sites and retrogradely labelled cells in the medulla, sections through the medulla and the cerebellum, in at least two different planes for each, were reacted with tetramethylbenzidine; alternate sections were reacted with cobalt-enhanced diaminobenzidine. Labelled were 60–80% of cells in the ipsilateral and 15–25% of cells in the contralateral external cuneate nuclei, as well as 25–50% of cells in the ipsilateral and the contralateral cell groups f and x near the descending vestibular nucleus.Substantial contralateral, as well as ipsilateral cerebellar projections from external cuneate nuclei and cell group x may be related to development of forelimb dexterity in raccoons, since these nuclei mediate forelimb muscular sensibility. The numerical complementarity of ipsilaterally versus contralaterally projecting cells suggests that they represent two separate populations.
