A Reactive 1O2 - Responsive Combined Treatment System of Photodynamic and Chemotherapy for Cancer

Abstract: The development of reactive oxygen species (ROS)-responsive drug delivery and drug release has gradually attracted much attention in recent years as a promising therapeutic strategy. Singlet oxygen (1O2) as the major ROS species is widely used in photodynamic therapy (PDT) of cancer. In the present study, we introduce a combined treatment using ROS-sensitive thioketal (TK) linkage as a linker between upconversion nanoparticles (UNs)-based PDT and doxorubicin (DOX)-based chemotherapy. UNs can not only play a ro… Show more

“…They can control the activation of PSs by adjusting the distance of PS and quencher [154]. Many stimuli, such as external stimuli (e.g., light [155]) and internal environment of tumor (e.g., pH [135,139,156,157,158,159,160,161,162,163], enzyme [91,164], GSH [165,166,167,168], and ROS [169,170,171,172,173,174,175,176]), have been used as the keys to turn on the PSs by increasing the distance among the PSs.…”

“…Further, thioketal-based drug delivery systems are widely reported in ROS triggered drug release 111 and in hybridized PDT-chemotherapy studies. 112 The stability of the thioketal bond under physiological conditions and its triggered cleavage only in the presence of ROS species could help to reduce off-target release of the cytotoxic drug. Thus, such properties make the thioketal linker a good choice for PDT-based chemotherapy.…”

“…The combination of 1 O 2 -responsive nanocarriers with other treatment, such as photodynamic therapy (PDT), has remarkable synergistic therapeutic effects (Wang et al, 2016 ; Yang et al, 2016 ; Yu et al, 2016 ; Li et al, 2017b ; Liu et al, 2017a ). In PDT, non-toxic photosensitizers are activated by exogenous light of a specific wavelength to transfer their excited energy to the surrounding oxygen to produce ROS.…”
