A region of the polyoma virus genome between the replication origin and late protein coding sequences is required in cis for both early gene expression and viral DNA replication

Abstract: Deletion mutants within the Py DNA region between the replication origin and the beginning of late protein coding sequences have been constructed and analysed for viability, early gene expression and viral DNA replication. Assay of replicative competence was facilitated by the use of Py transformed mouse cells (COP lines) which express functional large T-protein but contain no free viral DNA. Viable mutants defined three new nonessential regions of the genome. Certain deletions spanning the PvuII site at nt 51… Show more

“…Viral enhancers were originally identified in SV40 and polyoma virus as "far-upstream" promoter elements required for efficient early transcription (Benoist and Chambon, 1981;Gruss et al, 1981;Fromm and Berg, 1982;Tyndall et al, 1981). More recently, enhancer elements have been found in a variety of other viruses (reviewed in Gluzman and Shenk, 1983;Gruss, 1984;Picard, 1985).…”

“…Several reports about proteins interacting with enhancer sequences have been published recently (16,17,18,19,20,21). The enhancer of polyoma virus that is studied here is necessary both for transcription from the early promoter and for the replication of viral DNA (22,23,24). It could be subdivided into two fragments conserving partial activity: the PvuII-BclI fragment or A enhancer and the PvuII-4 fragment or B enhancer (25).…”

“…Recombinants carrying polyoma virus DNA produced slightly higher numbers of colonies compared to vector DNA alone. This is most probably due to the effect of polyoma virus enhancer elements (De Villiers & Schaffner, 1981 ;Tyndall et al, 1981 ;Herbomel et al, 1984) present in these constructs which function only weakly in the Chinese hamster cells. Since transformation frequencies correlate with enhancer activity in so many systems (Spandidos & Wilkie, 1983, 1984b and references therein), we take this to be a measure of enhancer function.…”
