A Review of Applications of Metabolomics in Cancer

Abstract: Cancer is a devastating disease that alters the metabolism of a cell and the surrounding milieu. Metabolomics is a growing and powerful technology capable of detecting hundreds to thousands of metabolites in tissues and biofluids. The recent advances in metabolomics technologies have enabled a deeper investigation into the metabolism of cancer and a better understanding of how cancer cells use glycolysis, known as the “Warburg effect,” advantageously to produce the amino acids, nucleotides and lipids necessary… Show more

“…In this way, it will be possible to monitor and discover metabolic changes related to disease onset or therapeutic interventions. Those techniques have been already applied to chronic renal diseases, hepatocellular carcinoma and colorectal cancers 185 as well as to oral cancer and periodontal diseases.…”

“…Thus, potential biomarkers that can address these issues relating to drug resistance and can predict the response of a patient to pemetrexed plus platinum doublet therapy are needed. Author Manuscript Published OnlineFirst on February 6, 2018; DOI: 10.1158/1078-0432.CCR- Metabolomics is an -omics technology that provides information that complements the genomic and proteomic profile of a sample (14). Metabolites are stable in serum and can be quantified, which presents an opportunity to establish a non-invasive approach for monitoring disease status and exploring biomarkers to predict the efficacy of anticancer therapies (15).…”

“…Recent research in this field has focused on metabolomics profiling for biomarkers discovery using different biological samples (tissue, urine, exhaled breath condensate, or plasma) [26]. However, as mentioned before, the complexity of serum/plasma and urine matrices is a factor to be considered in the search for biomarkers since other pathological conditions could interfere, particularly when a panel of markers formed by metabolites involved in primary metabolism is found.…”

“…This is the case with amino acids or compounds involved in energy metabolism (glycolysis or Krebs' cycle) [27] as they have shown to be modified in tumor cells. Specifically, potential markers found in urine included modified nucleosides, sarcosine, creatinine, or acetylglutamine [26,28]. However, the measurement of modified nucleosides levels have not shown to be reproducible or robust and sarcosine has also shown to fail in prostate cancer detection [29].…”
