A Role for IOP1 in Mammalian Cytosolic Iron-Sulfur Protein Biogenesis

Abstract: The biogenesis of cytosolic iron-sulfur (Fe-S) proteins in mammalian cells is poorly understood. In Saccharomyces cerevisiae, there is a pathway dedicated to cytosolic Fe-S protein maturation that involves several essential proteins. One of these is Nar1, which intriguingly is homologous to iron-only hydrogenases, ancient enzymes that catalyze the formation of hydrogen gas in anaerobic bacteria. There are two orthologues of Nar1 in mammalian cells, iron-only hydrogenase-like protein 1 (IOP1) and IOP2 (also kno… Show more

“…Current evidence indicates an important role for human Nbp35 in cytosolic iron-sulfur protein maturation (11). We have recently provided evidence that the human homologue of Nar1, IOP1, plays a role in cytosolic iron-sulfur protein maturation (12). IOP1 is homologous to iron-only hydrogenases, enzymes found in anaerobic bacteria that generate hydrogen gas from reducing equivalents.…”

“…In fact, seven mammalian relatives of yeast ISC and CIA proteins have been experimentally studied to date and shown to serve a similar function in the cell as their yeast counterparts [17][18][19][20][21][22][23] . The crucial importance of Fe/S proteins and their assembly machineries has been recognized by virtue of the association of lesions in ISC machinery genes with three human diseases (for a review, see refs.…”

“…IOP1 was first assumed to modulate the activity of hypoxia-inducible factor 1R that represents the global mediator of the mammalian response to hypoxia. Recently, depletion of the protein was shown to be essential for the formation of cytosolic Fe/S proteins identifying IOP1 as a functional Nar1 orthologue (18). Depletion of IOP2 (also termed NARF) was without effects on these Fe/S proteins, suggesting an alternative cellular function.…”
